Sounds relevant to me for a content role. They're likely trying to gauge your editorial aptitude and the depth of your knowledge with punctuation. People definitely have opinions about commas. Easy way to answer would be to limit it to the Oxford comma debate, whether you are pro or con. Or you could comment on how you notice their misuse (because that also happens all the time).

OMG - 20 years experience and they couldn't tell from your resume how your writing skills were?  Sounds like they had someone for the position already but had to go through the motions.  I went through the job description of the position I'm applying for and found several errors in grammar and punctuation.
